Lyophilised peptides are sensitive to moisture, static charge, and thermal variation. This protocol outlines the minimum requirements for safe and consistent handling in a laboratory environment.
Working Environment
Handle lyophilised peptide compounds in a clean, low-humidity environment. Ideally use a laminar flow cabinet or biosafety cabinet where available. Ensure the workspace is free from drafts which may cause powder displacement or contamination.Equipment Required
Calibrated micropipettes and compatible tips
- Pre-labelled cryogenic vials (1.5ml or 2ml)
- Analytical balance (0.1mg resolution minimum)
- Cryogenic storage rack
- Nitrile gloves, eye protection, lab coat
- Procedure
Allow sealed vials to equilibrate to room temperature before opening — this prevents condensation forming on the lyophilised cake.
Open vials carefully. The lyophilised cake may be fragile — avoid agitation.
Add reconstitution solvent (see Reconstitution Protocol) before aliquoting where possible. Aliquoting reconstituted solution is more accurate than dividing powder directly.
Where powder division is required, use a calibrated analytical balance in a draft-free environment. Transfer using a clean spatula — avoid metal contact with reactive compounds.
Transfer aliquots into pre-labelled cryogenic vials immediately.
Label each vial with: compound name, batch number, concentration, date of preparation, and initials of the preparing researcher.
Seal vials immediately after filling and transfer to -80°C storage.
Labelling Standards
All aliquots must be traceable to the original batch COA. Labels should be cryo-resistant and include: compound identity, lot/batch number, concentration, solvent used, preparation date, and storage conditions.Quality Notes
Discard any aliquots where powder colour, texture, or cake structure deviates from the reference COA description. Document discards in the laboratory inventory log.
